RV can be run on hosts that happen to use virtual IP address.
Basically, if you can ping the machine using its virtual IP address from some other machine in the subnet and it responds with its virtual IP address, then RV can work in this environment. If ping responds only with the physical address and not with the virtual address of the machine, then RV cannot operate correctly in such an environment.
In addition, if the IP address of a host that is running RVD is changed, then the RVD needs to be restarted in order to resolve/use the new IP address; the reason being the TIBCO Rendezvous daemon obtains interface information only at startup. In other words, so long as you shut down all of your RV applications and the RVD, while you make changes to the virtual IP and then once changes are completed you restart your applications and RVD, you shouldn't have any problem.
When you have more than one network interface, you should (generally speaking) specify the network parameter. If the interface you wish to use is default interface, then you don't have to. Though, in order to guard your application from being affected when someone changes which interface is the default interface, our recommendation would be to always specify network parameter when there are more than one interface on server.
